New paper on Small Angle Scattering out on Journal of Pharmaceutical Sciences!

Excelsus Structural Solutions'team is happy to announce the publication of the article 'Small Angle X-Ray Scattering Data Analysis and Theoretical Modelling for the Size and Shape Characterization of Drug Delivery Systems Based on Vitamin E TPGS Micelles' on Journal of Pharmaceutical Sciences. This work is the result of a collaboration with the Institute of Crystallography in Bari, Sapienza University of Rome and Novartis Pharma AG in Basel.

The article reports the development of a simple two-dimensional/two-components theoretical model that describes the structure and functionality of a VitE-TPGS system of micelles assuming a hydrophobic inner core and an outer hydrated hydrophilic shell. We then conceptually applied the developed methodology to a simple system of VitE-TPGS micelles unloaded and loaded with an active pharmaceutical ingredient, eltrombopag, to verify if the model could reliably monitor the size change of the micelle upon loading. The fit of laboratory Small Angle X-Ray Scattering data against such model allows us to extract absolute values of the micelles size under a spherical shape hypothesis as well as the distribution within the system between components and level of hydration.

Outstanding S-XRPD data collected on protein powders

In collaboration with the group lead by Professor Irene Margiolaki in Patras, Greece, Excelsus Structural Solutions‘ team recently published a breakthrough article on the application of high-resolution and high-statistics synchrotron X-Ray Powder Diffraction technique to protein powders. 

One of the major bottlenecks in macromolecular X-ray powder diffraction has always been the loss of structural information due to the severe overlap of powder diffraction peaks. The article 'New perspectives in macromolecular powder diffraction using single photon counting strip detectors: High resolution structure of the pharmaceutical peptide Octreotide' published in Acta Crystallographica Section A, describes an improved data-collection strategy using a MYTHEN II strip detector that leads to substantial improvement of angular resolution at low 2θ. The application of this strategy enabled the project team to achieve a 1.8 Ang structural model of the molecule "octreotide", surpassing the model achieved in the past.

Excelsus cited in the new "Mediator" section of the EARIV website

In the context of the CAROTS EU-project, existing Mediators companies have been identified and linked to form a Mediator network. Mediators, such as Excelsus Structural Solutions are commercially oriented entities specialized in servicing industrial companies with analysis, measurement services and experiments at synchrotrons and neutron facilities. A new Mediator section has recently been published on the EARIV website.

Presentation of ESS Case studies at the joint CCG/IG Autumn Meeting
20th November 2019

The joint CCG/IG autumn meeting "By hook or by crook - structural solutions from challenging crystals" was held at GSK site in Stevenage on the 20th November 2019. On this occasion, Mathilde Reinle-Schmitt presented results collected in the frame of ESS Case Studies program, launched for the joint SPS-XRPD-2/PPXRD-16 conference in May 2019. The talk was entitled 'Beyond the limits of conventional techniques: subtle structural differences influencing solubility unveiled by synchrotron-XRPD' and described how the use of S-XRPD could solve issues in industrial pharmaceutical R&D.
